Buck ultimately succumbed to a lifetime of smoking. He died of lung cancer at M.D. Anderson Hospital, in Houston, on March 25, 1950, at age 66. He was cremated and his ashes were scattered, as was reported, "in Texas". He was a native of Gainesville, Texas, where the zoo was named for him, and spent his last years at the family home in San Angelo.

Muriel Reilly was Buck's 3rd wife, from 1928 until his death in 1950. With her he had his only child, a daughter, Barbara.
